RNA sequencing data for gamma radiation response in the extremotolerant tardigrade Ramazzottius varieornatus

چکیده

Tardigrades are microscopic animals of which terrestrial species capable tolerating extreme environments by entering a desiccated ametabolic state known as anhydrobiosis. Intriguingly, they survive high dosage gamma rays (>4,000 Gy), possibly through mechanism cross-tolerance. We hypothesized that anhydrobiosis genes also regulated during cross-tolerance, thus we submitted Ramazzottius varieornatus to 500 Gy 60 Co gamma-ray and conducted time-course low-input RNA-Seq. The gene expression was quantified with RSEM differential determined DEseq2. Differentially expressed were ontology enrichment analysis GOStat. transcriptome dynamically shifted nine hours post-exposure.

Analysis of DNA Repair and Protection in the Tardigrade Ramazzottius varieornatus and Hypsibius dujardini after Exposure to UVC Radiation

Tardigrades inhabiting terrestrial environments exhibit extraordinary resistance to ionizing radiation and UV radiation although little is known about the mechanisms underlying the resistance. We found that the terrestrial tardigrade Ramazzottius varieornatus is able to tolerate massive doses of UVC irradiation by both being protected from forming UVC-induced thymine dimers in DNA in a desiccat...

Comparative genomics of the tardigrades Hypsibius dujardini and Ramazzottius varieornatus

Tardigrada, a phylum of meiofaunal organisms, have been at the center of discussions of the evolution of Metazoa, the biology of survival in extreme environments, and the role of horizontal gene transfer in animal evolution. Tardigrada are placed as sisters to Arthropoda and Onychophora (velvet worms) in the superphylum Panarthropoda by morphological analyses, but many molecular phylogenies fai...

Extremotolerant tardigrade genome and improved radiotolerance of human cultured cells by tardigrade-unique protein

Tardigrades, also known as water bears, are small aquatic animals. Some tardigrade species tolerate almost complete dehydration and exhibit extraordinary tolerance to various physical extremes in the dehydrated state. Here we determine a high-quality genome sequence of Ramazzottius varieornatus, one of the most stress-tolerant tardigrade species. Precise gene repertoire analyses reveal the pres...

Novel Mitochondria-Targeted Heat-Soluble Proteins Identified in the Anhydrobiotic Tardigrade Improve Osmotic Tolerance of Human Cells

Tardigrades are able to tolerate almost complete dehydration through transition to a metabolically inactive state, called "anhydrobiosis". Late Embryogenesis Abundant (LEA) proteins are heat-soluble proteins involved in the desiccation tolerance of many anhydrobiotic organisms. Tardigrades, Ramazzottius varieornatus, however, express predominantly tardigrade-unique heat-soluble proteins: CAHS (...

A Graph-Based Clustering Approach to Identify Cell Populations in Single-Cell RNA Sequencing Data

Introduction: The emergence of single-cell RNA-sequencing (scRNA-seq) technology has provided new information about the structure of cells, and provided data with very high resolution of the expression of different genes for each cell at a single time. One of the main uses of scRNA-seq is data clustering based on expressed genes, which sometimes leads to the detection of rare cell populations. ...
